Small RNAs are noncoding RNA molecules < 200 nt in length. Eukaryotic small RNAs are classified into biotypes based on their molecular characteristics and fundamental biological functions: microRNAs (miRNA), PIWI-interacting RNAs (piRNA), short interfering RNAs (siRNA), small nucleolar RNAs (snoRNA), small nuclear RNAs (snRNA), transfer RNAs (tRNA), and tRNA-related fragments and halves (tRF) (Figure 1). Some types of small RNAs perform fundamental functions, such as tRNA in protein translation and snoRNA in rRNA modification. Some others are regarded as small regulatory RNAs which regulate gene expression and modulate biological activities by multiple mechanisms, such as mRNA stability and translation control, or targeting epigenetic modifications to specific genomic regions [1-3]. Their stability, enrichment abundance in biofluids have been utilized for biomarker applications.

Small RNA biogenesis pathways are the underlying processes that produce these small RNAs and are regulated in response to cellular conditions. Disorders of the pathways are often associated with diseases. For example, the expression levels of DROSHA and DICER1, the key protein factors in miRNA biogenesis, change along with the stages of tumor progression, which have been investigated as a potential biomarker for neuroblastoma prognosis [4]. Profiling small RNAs biogenesis pathways facilitates the study of the small RNAs, the biological impacts and the associated diseases.
